Composition

Each 2 ml pre-filled syringe contains:

• Polynucleotides PN-HPT™ — 10 mg/ml
• Non-cross-linked sodium hyaluronate — 10 mg/ml
• Mannitol
• Excipients: Aqua, Sodium Phosphate, Disodium Phosphate

PN-HPT™ (Polynucleotide Highly Purified Technology) refers to DNA fragments derived from highly purified Italian trout (Oncorhynchus mykiss) using Mastelli's patented extraction process. The purification level meets pharmaceutical-grade standards, minimising immunogenic risk. Non-cross-linked hyaluronic acid provides immediate dermal hydration without volumising effect. Mannitol acts as a free-radical scavenger, extending HA activity and supporting the antioxidant component of treatment.

Mechanism of Action

The three active components work synergistically across distinct biological pathways.

Polynucleotides support fibroblast proliferation and metabolic activity, supporting collagen and elastin synthesis and extracellular matrix remodelling. Non-cross-linked HA restores hydration in the dermis by binding water molecules, improving skin turgor and suppleness. Mannitol neutralises reactive oxygen species, protecting dermal structures from photo- and chrono-induced oxidative damage and extending HA activity.

Clinical Indications

Mastelli Newest is indicated for biorevitalisation of:

• Face (including periorbital and perioral areas)
• Neck and décolleté
• Hands
• Areas affected by atrophic (concave) scars and stretch marks

Injection Technique

• Depth: intradermal
• Techniques: linear, retrograde, point-by-point, cross-hatching, fan, or combined
• Alternative delivery: iontophoresis or electroporation
• Post-injection: gentle massage to distribute the product evenly

Treatment Planning

Initial series: 3 sessions at 3-week intervals. Maintenance: single session every 3 months. More intensive scheduling may be considered during periods of elevated UV exposure.

Results and Duration

Improvement in skin texture, hydration and firmness is typically observable from the first session. Results are reported to last 4–6 months depending on age, skin condition and lifestyle. Photoprotection is recommended throughout the treatment period.
